About this deal

Therapeutically given folic acid is converted into the metabolically active form 5MTHF in the plasma and liver. There is an enterohepatic circulation for folate. Non-drug - induced folic acid deficiency, or abnormal folate metabolism, is related to the occurrence of birth defects and some neural tube defects. Variation in response to folates as well as other xenobiotics is multifactorial, and includes genetic and environmental factors. Evaluation of genetic contribution typically comes from clinical observations, while the contribution of environmental factors is difficult to control and manipulate in the living environment of clinical subjects. These limitations can be overcome by applying cell-based pharmacogenomic models, such as the lymphoblastoid cell lines (LCLs). LCLs are generated from fresh lymphocytes isolated from blood samples donated by consenting healthy adults and transformed by the Epstein-Barr virus (EBV); therefore, they can be grown in culture. Their nuclear DNA remains intact after immortalization; consequently, LCLs are a preferred method of storing an individual’s genetic material in biobanking [ 31]. These cell lines display the full spectrum of enzymes of the folate-homocysteine metabolism pathway and their reactions, and are a suitable model for studying metabolites of the folate-homocysteine cycle. LCL models have been widely used for forward and reverse translational research, where phenotypic observation or clinical findings are validated by functional LCL follow-up studies aiming to understand the mechanism of action. LCLs provide a cost-effective testing system where environmental factors and dosage can be controlled. Using LCL derived from diverse individuals makes it possible to mimic population diversity. Both vitamin B12 deficiency and folate deficiency are more common in older people, affecting around 1 in 10 people aged 75 or over and 1 in 20 people aged 65 to 74. Treating vitamin B12 or folate deficiency anaemia If you have a higher chance of having a baby with a neural tube defect your doctor will recommended an increased dose of 5mg, taken once a day. Dose for treating folate deficiency anaemia

You'll usually take it for 4 months. But if your folate deficiency anaemia is caused by a long-term problem, you may have to take folic acid for longer, possibly for the rest of your life. Do not stop taking folic acid without talking to your doctor. Dose for preventing folate deficiency anaemiahelp your baby's brain, skull and spinal cord develop properly in pregnancy, to avoid development problems (called neural tube defects) such as spina bifida

A meta-analysis of 26 randomized controlled trials found that folic acid supplementation was not associated with a decreased risk of cardiovascular disease, heart disease, or deaths from these conditions, but it was linked with a decreasing trend in risk of strokes. [21] When 6.25 nM, 12.5 nM, 25 nM, and 50 nM 5-Me-THF were added into folate-deficient medium, the proportion of cells in the S-phase decreased to 16.1% ( p = 0.038), 14.5% ( p = 0.0032), 12.9% ( p = 0.0002), and 12.0% ( p< 0.0001), respectively ( Figure 2D). When the equimolar concentrations of FA were added, the proportions of cells in the S-phase were 18.9% (n.s.; p = 0.38), 17.0% ( p = 0.024), 16.1% ( p = 0.0042), and 13.7% ( p< 0.0001), respectively ( Figure 2C). Your doctor or midwife may recommend taking folic acid throughout pregnancy, particularly if you are at risk of anaemia or are anaemic. Supplementation with active folate 5-MTHF bypasses the entire folate metabolization, which is potentially impaired by MTHFR polymorphism, and 5-MTHF is directly absorbed to exert the biological activity. Therefore, using 5-MTHF as a food supplement instead of FA is strongly recommended for external supplementation. 13 Folate is the generic term for vitamin B9, a water-soluble vitamin that includes chemically similar compounds essential in periods of rapid cell growth and division, in the maintenance of new cells, and in the making of DNA and RNA. While best recognized for pregnancy and fertility support, recognition is growing that it also benefits cardiovascular health, mood, and cognition.

Ericson U, Sonestedt E, Gullberg B, Olsson H, Wirfält E. High folate intake is associated with lower breast cancer incidence in postmenopausal women in the Malmö Diet and Cancer cohort. The American journal of clinical nutrition. 2007 Aug 1;86(2):434-43. The differences in MA of cells treated with 5-Me-THF compared to FA at 12.5 nM, 25 nM, and 50 nM were 12% ( p = 0.031), 15% ( p = 0.0071), and 11% ( p = 0.048), respectively. At 6.25 nM and 100 nM, the difference between MA of LCLs treated with FA or 5-Me-THF was not evident ( p = 0.18, p = 0.97). A 2014 published study clearly showed that 86% of FA in the hepatic portal vein is unmetabolized, while almost all the natural folate was converted correctly. 19, 20 Detectable levels of UMFA occur temporarily in plasma after the consumption of >200 μg FA, with concentrations increasing in parallel to that of total FA after supplementation. UMFA has been detected in cord and infant blood, a source of concern due to potential adverse effects on health, as I will further describe. 18, 21
